ABSTRACT

Objective: To investigate the value of ultra-high b value diffusion-weighted imaging (DWI) and intravoxel irrelevant motion (IVIM) imaging in differential diagnosis of prostate cancer (PCa) and benign prostate hyperplasia (BPH) and predicting pathological grade of PCa. Methods: Totally 55 patients who underwent prostate IVIM-DWI (b=0, 50, 100, 150, 200, 400, 600, 800, 1 000, 1 500, 2 000 s/mm2) and were pathologically confirmed as PCa (PCa group) or BPH (BPH group) were collected. Patients in PCa group were divided into low score subgroup or high score subgroup according to the pathological grading. The differences of ADC value, D value, D* value and f value were compared between PCa and BPH groups as well as low score subgroup and high score subgroup. ROC curve for differential diagnosis of PCa and BPH and predicting PCa pathological grading were drawn to evaluate the diagnostic efficacy of the indexes significantly different between PCa and BPH or low and high score subgroups of PCa, and the correlation between parameters and Gleason scores in PCa group were analyzed. Results: A total of 44 patients were enrolled, including 24 cases in PCa group and 20 in BPH group. ADC value and D value were statistically different (t=-14.24, -10.87, both P<0.01), while AUC of ADC value (0.99) and D value (0.98) for differentiating PCa and BPH were not between 2 groups (Z=1.18, P=0.24). In PCa group, ADC value and D value were statistically different (t=2.28, 3.86, both P<0.05), and AUC of ADC value (0.76) and D value (0.88) for predicting pathological grading of PCa were also statistically different between 2 subgroups (Z=2.23, P=0.02). ADC value (r=-0.44)and D value (r=-0.56) were both negatively correlated with Gleason score (both P<0.05) in PCa gruop. Conclusion: Ultra-high b value DWI and IVIM imaging had certain application value in PCa. For differential diagnosis of PCa and BPH, the efficiency of ADC value was comparable to that of D value, while for predicting pathological grading of PCa, the efficiency of D value was better than that of ADC value.

ABSTRACT

OBJECTIVE Alzheimer disease(AD),the most common cause of dementia among older people, could not be prevented, halted, or reversed up till now. A large body of pharmacological study has revealed that Liuwei Dihuang (LW) possesses potential therapeutic effects on AD. LW-AFC is key fractions from LW.In the present study,we investigated the effect of LW-AFC on AD mouse models. METHODS PrP-hAβPPswe/PS1ΔE9(APP/PS1) mice and senescence-accelerated mouse prone 8 strain (SAMP8), classic AD animal models, were employed. After the treatment of LW-AFC, mice were cognitively evaluated in behavioral experiments. Neuron loss, amyloid-β (Αβ) deposition, and Αβ level were analyzed using Nissl staining, immunofluorescence, and an AlphaLISA assay, respectively. Multiplex bead analysis, a radioimmunoassay, immunochemiluminometry, and an ELISA were used to measure cytokine and hormone levels.Lymphocyte subsets were detected using fl ow cytometry. RESULTS LW-AFC ameliorated the cognitive impairment observed in APP/PS1and SAMP8 mice,including the impairment of object recognition memory,spatial learning and memory,and active and passive avoidance. In addition, LW-AFC alleviated the neuron loss in the hippocampus, suppressed amyloid-β(Αβ)deposition in the brain,and reduced the concentration of Aβ1-42in the hippo-campus and plasma of APP/PS1 mice. LW-AFC treatment also significantly restored the imbalance of hypothalamic-pituitary-adrenal (HPA) and hypothalamic-pituitary-gonadal (HPG) axis, enhanced the proliferation of splenocytes,corrected the disorder of lymphocyte subsets,and regulated the abnormal production of cytokine in APP/PS1 and SAMP8 mice. Effects of LW-AFC on pharmacodynamics and neuroendocrine immunomodulation network in APP/PS1 and SAMP8 mice were better than meman-tine and donepezil. CONCLUSION LW-AFC ameliorated the behavioral and pathological deterioration of AD mouse models via the restoration of the NIM network, which supports the use of LW-AFC as a potential agent for AD therapy.

ABSTRACT

Technologies for glycomics usually involve methods for separation and purification of poly-saccharides, and separation, structure resolution, quantification, property investigation and function comment of glycan chains. Because of the different biochemical properties of glycoproteins, proteogly-cans and glycolipids, the separation and purification of polysaccharides involve corresponding fractional precipitation, boric acid affinity, titanium dioxide, affinity chromatography, size exclusion method, and gel filtration chromatography column chromatography methods. The lectins, water affinity chromatogra-phy , solid phase extraction and other technologies could be applied to the oil enrichment of high pure and specific glycan chains. The structure of glycan chains can be analyzed using lectin microarray technolo-gy, mass spectrometry, and derivatization markers of glycan chains. lsotope labelling and metabolic labeling can be used to quantify glycan chains. The glycan biological function can be better understood using glycan chain structure analysis software and database of glycan chains by bioinformatics.
